PRINCIPLE

 

The Strep A Rapid Test is a qualitative, lateral flow immunoassay for the detection of Strep A carbohydrate antigen in a throat swab. In this test, antibody specific to Strep A carbohydrate antigen is coated on the test line region of the test. During testing, the extracted throat swab specimen reacts with an antibody to Strep A that is coated onto particles. The mixture migrates up the membrane to react with the antibody to Strep A on the membrane and generate a color line in the test line region. The presence of this color line in the test line region indicates a positive result, while its absence indicates a negative result. To serve as a procedural control, a colored line will always appear in the control line region, indicating that proper volume of specimen has been added and membrane wicking has occurred.

 

PROCEDURE

 

Bring test devices, specimens, and/or controls to room temperature (15- 30°C) before use.

1. Remove the test device from its sealed pouch, and place it on a clean, level surface. Label the device with patient or control identification. For best results, the assay should be performed within one hour.

2. Gently mix Extraction reagent solution. Remove the foil, place the patient swab specimen into the Extraction Tube with buffer.

3. Roll the swab at least 10 times while pressing the swab against the bottom and side of the Extraction Tube. Roll the swab head against the inside of the Extraction Tube as you remove it. Try to release as much liquid as possible. Dispose of the used swab in accordance with your biohazard waste disposal protocol.

4. Put on the tube tip, then add 3 drops (100μL) of extracted sample into each sample well. Do not handle or move the Test Device until the test is complete and ready for reading. As the test begins to work, color will migrate across the membrane. Wait for the colored band(s) to appear. The result should be read at 15 minutes. Do not interpret the result after 20 minutes.
